1. Principle

This AIV-H5 antibody rapid test card is formulated based on the principle of gold immunochromatography assay (GICA). After adding the sample to the loading well, it moves along the chromatographic membrane together with the colloidal gold marker. If there is H5 subtype avian influenza antibody in the sample, it will bind to the colloidal gold marker and the antigen on the testing line, resulting in a purple-red color. If there is no H5 subtype avian influenza antibody in the sample, no color reaction will occur.

3. Specimen requirements

1) This test card is only suitable for testing avian serum/whole blood.

2) Serum: collect 2-3ml of blood according to conventional methods and place it in a clean and dry test tube. Let it stand for about 1 hour until the blood coagulates, and then centrifuge it at 4000 rpm for 10 minutes (alternatively, let the blood stand for about 2 hours until the serum naturally separates out after blood coagulation). Separate the serum. The serum should be clear, without hemolysis or contamination. Serum samples can be stored at 2-8℃ for short-term storage, and at -20℃ for long-term storage.

3) Whole blood: Collect fresh anticoagulant-treated whole blood for immediate use. Collect and use whole blood without anticoagulant immediately.

4. Test procedure

1) Serum/whole blood sample addition method: Tear the aluminum foil packaging bag of the test card, take out the test card, and place it on a flat and clean surface. Use the dropper inside the bag to take 1 drop of serum or 2 drops of whole blood and slowly dispense it into the sample well. Then add 2 drops of AIV-H5 Ab Sample dilution to the sample well.

2) Place it at room temperature and judge the result after 10-15 minutes. The color development result after 15 minutes is invalid.

5. Results judgment

1. Negative: only control line is seen purple-red red;

2) Positive: control line and test line are both seen wine red. The higher the antibody titer, the darker the color of the test line (T).

3) Invalidation: control line isn’t seen wine red.

The detection threshold of this test kit is HI test 1:32. Therefore, by diluting the tested serum exponentially and finding the maximum dilution that shows positive results, the antibody titer can be determined. For example, dilute the serum to be tested with physiological saline at 1:2, 1:4, and 1:8, respectively. Then test the three samples with different dilution ratios. If both 1:2 and 1:4 show positive results, while 1:8 shows negative results, the maximum dilution that shows positive results is 1:4. Therefore, the final antibody titer of this sample is 2⁵ (1:32) multiplied by 2² (1:4), which equals 2⁷ (1:128).

6. Explanation of testing results

For samples post-immunization, the level of antibody indicates the effectiveness of the immunization. For unimmunized samples, a positive result suggests potential infection with H5 subtype avian influenza virus, which requires analysis in conjunction with clinical and other methods.

7. Limitations of the testing method

This test card is only for screening and is not used as a basis for diagnosis.

8. Precautions

1). Please read the instruction manual carefully before the test. The sample dilution provided by this product are solely for use in this test.

2). Please do not use other animal serum or any other non-【Specimen requirements】 liquid as a negative control.

3). Products that have expired or whose aluminum foil bags are damaged are not to be used.

4). The test card should be allowed to return to room temperature before being opened after being taken out of the refrigerator. The opened test card should be used as soon as possible to avoid becoming ineffective due to moisture.

5). Avoid using samples that are turbid, have severe hemolysis, or contain a large amount of lipids, as these may contaminate the test.

6). Avoid touching the chromatographic membrane at the sample application well and detection window on the test card.

7). Waste from the experiment should be regarded as pollutants and disposed of properly in accordance with local regulations.
