theChloramphenicol(CAP) rapid test dipsticks (Egg)
Catalog No. LSY-20004JD
1. Brief
This product is used for testing theChloramphenicol in egg, duck egg sample qualitatively. The whole test need 30-45min. Easy to operate, high sensitivity.
2. Detection limit
Egg: 0.1μg/kg(ppb)
3. Contents
1 | CAP test strip and micro-well reagent | 8 strip/bottle, 12 bottle/kit |
2 | Instructions | 1 piece |
3 | Reagent 1 | 2 bottles |
4 | Reagent 2 | 2 bottles |
5 | 10X concentrated sample buffer | 1 bottle |
4. Principle
The colloidal gold immuno-chromatography was used to detect the theChloramphenicol residue in the samples. The binding of theChloramphenicol to the specific monoclonal antibody labeled with colloidal gold inhibited the binding of the antibody to the conjugates of theChloramphenicol-BSA on the NC membrane Testing line, which resulted in the color depth variation of the testing line.
5. Testing equipment required
Instruments: balance, centrifuge, sample concentrator, micropipette, oscillator (optional), homogenizer
Consumables: 50ml centrifuge tube, disposable tips, ethyl acetate, n-hexane.
6. Sample preparation
1) Sample buffer: Dilute 10X concentrated sample buffer with deionized water at 1:9 to get sample buffer. (for example: 1 part of 10X concentrated sample buffer + 9 parts of deionized water)
2) Egg: Take 3±0.05g fresh and evenly mixed egg samples into 50ml centrifuge tube, add 1ml of Reagent 1, shake by hand or oscillator for 2min, add 1ml of Reagent 2, then add 7ml ethyl acetate, shake by hand or oscillator for 2min, centrifuge at 4000r/min for 5min; take 4ml supernatant into another centrifuge tube, blow to dry at 56℃ by Nitrogen or air (or use hair dryer); add 2ml of n-hexane, shake for 30S to dissolve the solid on wall and bottom of tube fully, then add 0.45ml, shake for 10S, be static or centrifuge at 4000r/min for 5min, until liquid layered, discard up-layer liquid, absorb 200ul of down-layer liquid, ready for testing.
7. Operation procedures
7.1 Read the instruction carefully before use. Return test strips and sample into room temperature.
7.2 Take bottles needed from the kit package, take out required microwells and dipsticks, making proper marks. Please use these test dipsticks within 1h. Seal the cap of the bottles, avoid moisture.
7.3 Take 200ul of the test samples into the microwells, then slowly aspirate for 5 times, mix the sample with the reagent in the microwells completely, until no solid observed by eyes (this is a very important step).
7.4 Incubate for 5min at room temperature (20-25℃), then insert the test strips into the microwells with the "MAX" end fully dipped in to the mixture solution.
7.5 After immersing the test strip in the micro-well for 5 minutes, interpret the results according to the schematic diagram. Please interpret the results within 1 minutes, otherwise interpretation will be invalid.
8. Test Result Interpretation
8.1 Negative: T line is darker than C line, or T line has same color with C line. It means there is no theChloramphenicol residue in sample or the residue is lower than detection limit.
8.2 Positive: T line is obviously lighter than C line or T line is invisible. It means the theChloramphenicol residue is equal to or higher than detection limit.
8.3 Invalidation: C line isn’t seen wine red. It means the test strip is out of efficacy, out of date or improper operation. Please run the test again using another package. If the invalid tests keep happening, please contact the supplier.
9. Precautions
1) The test strips can be used only once at room temperature, do not use test strips out of expiry date.
2) Please seal the bottle after taking out required test strips. If can not use all 8 micro-wells at once, cup the rest and put it back to bottle and sealed.
3) Do not touch the white membrane surface in the middle of test strips, avoid sunlight and fan blowing directly.
4) The egg sample must be a uniform liquid, without clumping, souring, or precipitation.
5) Use the test strips testing again for positive results.
6) Please contact the supplier for any questions.
7) The result is for reference only, for confirmation, please refer to the relevant national standard method.
10. Safety performance and garbage disposal
This product's test strips and packaging are non-toxic and harmless, and should be disposed of as dry waste after use; The organic reagents used are collected and centrally processed in waste liquid bottles.
11. Storage and expiry date
Storage: Store at 2-8 ℃ in dark, sealed, dry place, no frozen.
Expiry date: 12 months; date of production is on box.
