1. Brief
This product is used for testing Zearalenone in grain, feed qualitatively, the whole test procedure only need 10-15min, easy to operate with high sensitivity.
2. Detection limit: 60~1500μg/kg(μg/kg=ppb)

4. Principle
The Zearalenone(ZEN) rapid test strip is based on competitive inhibition immuno-chromatographic principle. In the flow process, Zearalenone in the sample combined with Zearalenone specific colloidal gold-labeled monoclonal antibody, Inhibit the combination between antibody and ZEN -BSA conjugate on Test line of NC membrane, lead to the color change of Test line.
When the sample has no Zearalenone residue or concentration lower than detection limit, T line is darker than or has same color with C line. when the concentration is equal to or higher than detection limit, T line is lighter than C line obviously or T line is invisible. No matter whether there is Zearalenone residue in sample, C line will appear, it means the test is valid.

6. Reagents and equipment required but not provided
6.1 Electronic balance (precision above 0.01g)
6.2 Pipette (10-100μL, 20-200μL, 100-1000μL, 1-10mL)
6.3 Centrifuge tube (50mL, 4mL, 2mL)
6.4 Ethanol (analytical grade), acetonitrile (analytical grade), pure water
6.5 Vortex meter, centrifuge, timer, etc.
7. Solution preparation
50% ethanol-water solution: 50mL ethanol (analytical purity) + 50mL pure water
8. Sample preparation
Weigh 5.0±0.05g homogenous (passing 20 mesh screen) sample into a 50mL centrifuge tube, add the corresponding extraction solution as shown in the table below, with a stopper, fully shake for 3min, centrifuge at 4000r/min for 5min or stand still to separate, get the up-layer clear liquid.

9 Sample dilution
According to the following method, add the up-layer clear liquid and Zearalenone sample dilution, mix it evenly, and ready for testing.
1). 60-500μg/kg detection limit
Sample I: Corn, wheat, rice bran, bran, wheat flour and flour
Take 50μL of the up-layer clear liquid (100μL of the up-layer clear liquid for rice bran, bran and wheat flour samples) and add it to the Zearalenone sample dilution according to the following detection limits;

Take 50μL of up-layer clear liquid (100μL of up-layer clear liquid for corn gluten meal and other strongly absorbent samples), and add to the Zearalenone sample dilution according to the following detection limits;

Take 25μL of up-layer clear liquid (50μL of up-layer clear liquid for corn gluten meal and other strongly absorbent samples), and add to the Zearalenone sample dilution according to the following detection limits;

10. Operation procedures
10.1 Read the instruction carefully before use. Take tubes needed from the kit package, take out required micro-wells and strips, making proper marks.Seal the cap of the tubes, avoid moisture.
10.2 Return test strips,micro-wells and sample into room temperature(20~25℃)before use. Place the required micro-well reagents in the empty micro-well plate rack; do not exceed 8 samples at a time; if it exceeds, test in groups of 8 in batches.
10.3 Take 200ul of the test samples into the micro-wells, then repeatedly pump and suck for 5~6 times, mix the sample with the reagent in the micro-wells completely until no solid judge by eyes (this is a very important step). Use the timer to start timing.
10.4 Incubate for 3min at room temperature(20-25℃), then insert the test strips into the micro-wells with the "MAX" end fully dipped in to the mixture solution.
10.5 Insert the test dipsticks into micro-wells for 5min at room temperature(20-25℃), take out the test strip and remove the sample pad at the lower end and interpret the result within 1 min according to the picture. The interpretation at other times is invalid.
11. Test Result Interpretation
9.1 Negative: T line is darker than or has same color with C line. It means there is no Zearalenone residue in sample or the residue is lower than detection limit.
9.2 Positive: T line is obviously lighter than T line or T line is invisible. It means the Zearalenone residue is higher than or equal to detection limit.
9.3 Invalidation: C line isn’t seen wine red. It means the test strip is out of efficacy, out of date or improper operation. Please run the test again using another package. If the invalid tests keep happening, please contact the local distributor.
12. Precautions
1. Return the test strip and the sample to be tested to room temperature before use.
2. Please use the test strip within the validity period.
3. When opening the micro-wells reagent, please gently uncover the sealing film, do not uncover it quickly, so as to avoid powder flying out.
4. Please use up the micro-wells reagent as soon as possible after opening, otherwise it will become invalid and affect the detection effect.
5. Do not touch the white film surface in the center of the test strip.
6. Avoid direct sunlight and direct blowing of electric fans during testing.
7. Do not use tap water, purified water and distilled water as negative controls.
8. The up-layer clear liquid of corn sprouts, sprayed corn, corn gluten meal and some finished feeds is acidic or alkaline after extraction. When acidic pH <5 or alkaline pH> 9, Adjust the pH of the up-layer clear liquid to 6.0-8.0 with the 2.0 mol/L NaOH solution or 2.0mol/L HCL solution before diluting and testing.
9. Due to the complex formula of the finished feed, it may contain substances that interfere with detection; the results of negative spiked samples and real samples are slightly different due to different toxin binding methods.
10. This method is a screening method. If a positive sample or a suspected positive sample is encountered, use the confirmation method to confirm.
