Deoxynivalenol(DON) rapid test strip
1. Brief
This product is used for testing Deoxynivalenol in grain, feed qualitatively, the whole test procedure only need 10-15min, easy to operate with high sensitivity.
2. Detection limit: 1000~5000μg/kg (μg/kg=ppb)

4. Principle
The Deoxynivalenol(DON) rapid test strip is based on competitive inhibition immuno-chromatographic principle. In the flow process, Deoxynivalenol in the sample combined with Deoxynivalenol specific colloidal gold-labeled monoclonal antibody, Inhibit the combination between antibody and DON -BSA conjugate on Test line of NC membrane, lead to the color change of Test line.
When the sample has no Deoxynivalenol residue or concentration lower than detection limit, T line is darker than or has same color with C line. when the concentration is equal to or higher than detection limit, T line is lighter than C line obviously or T line is invisible. No matter whether there is Deoxynivalenol residue in sample, C line will appear, it means the test is valid.

6. Self prepared reagents and instruments
1). Electronic balance (accuracy above 0.01g)
2). Pipette (10-100 μL,20-200 μL,100-1000 μL,1-10mL)
3). Centrifuge tube (50mL, 2mL or 1.5mL)
4). Pure water
5). Vortex meter, centrifuge, timer, etc
7. Sample preparation
Weigh 5.0±0.05g homogeneous sample (passing through 20 mesh screen) into a 50mL centrifuge tube, add 20mL pure water into the centrifuge tube, fully shake for 3min, centrifuge at 4000r/min for 5min or leave it for layering to obtain the supernatant.
8. Sample dilution
Add supernatant and DON sample dilution according to the following method, and mix them well for inspection.
1) 1000μg/kg detection limit
Take 1000μL DON sample dilution, add into 2mL or 1.5mL centrifuge tube, then add 60μL supernatant, mix well and wait for test.
2) 3000μg/kg detection limit
Take 1000μL DON sample dilution, add into 2mL or 1.5mL centrifuge tube, then add 20μL supernatant, mix well and wait for test.
3) 5000μg/kg detection limit
Take 2000μL DON sample dilution, add into 2mL or 1.5mL centrifuge tube, then add 23μL supernatant, mix well and wait for test.
9. Operation procedures
9.1 Read the instruction carefully before use, take tubes needed from the kit package, take out required micro-well reagents and strips, making proper marks. Seal the tube immediately after take out the strips and reagents, avoid moisture.
9.2 Return test strips,micro-well reagents and samples into room temperature (20~25℃) before use, place the required micro-well reagents in the empty micro-well plate rack; The number of samples tested at one time shall not exceed 8; If it exceeds the limit, it can be tested in batches according to 8 samples per group.
9.3 Take 200ul of the diluted sample into the micro-well reagent, then slowly aspirate until the testing sample and micro-well reagent are fully mixed, about 5-6 times; use the timer to start timing for 3min.
9.4 After Incubate for 3min at room temperature(20-25℃), then insert the test strips into the micro-wells with the "MAX" end fully dipped in to the mixture solution, then start timing.
9.5 After Incubate for 5min at room temperature(20-25℃), read the result, If it is impossible to interpret immediately, the sample pad shall be stripped at the end of incubation to ensure the accuracy of delayed interpretation of experimental results.
10. Test Result Interpretation
10.1 Negative: T line is darker than or has same color with C line. It means there is no Deoxynivalenol residue in sample or the residue is lower than detection limit.
10.2 Positive: T line is obviously lighter than T line or T line is invisible. It means the Deoxynivalenol residue is higher than or equal to detection limit.
10.3 Invalidation: C line isn’t seen wine red. It means the test strip is out of efficacy, out of date or improper operation. Please run the test again using another package. If the invalid tests keep happening, please contact the local distributor.
11. Precautions
1). Restore the test strip, and sample to room temperature before use.
2). Please use the test strip within the validity period.
3). When opening the micro-well reagent, please gently open the sealing film, and do not use force to quickly open it, so as to avoid powder flying out.
4). Please use up the micro-well reagent as soon as possible after opening, otherwise it will become invalid and affect the detection effect.
5). Do not touch the white film surface in the center of the test strip.
6). Avoid direct sunlight and direct blowing of electric fan during detection.
7). Do not use tap water, purified water and distilled water as negative control.
8). Due to the complex formula of the finished feed, it may contain substances that interfere with the detection; The results of negative spiked samples and real samples were slightly different due to different toxin binding modes.
9). This method is a screening method. In case of positive samples or suspected positive samples, please confirm with confirmation method.
