2. Brief
This product is used for testing Leucomalachite Green in Tissue (Fish, shrimp, and crab: tissues of freshwater fish, marine fish, shrimp, crabs, etc) sample qualitatively. Easy to operate, high sensitivity.

5. Principle
The Leucomalachite green rapid test card is based on competitive inhibition immuno-chromatographic principle. In the flow process, Leucomalachite green in the sample combined with Leucomalachite green specific colloidal gold-labeled monoclonal antibody, Inhibit the combination between antibody and Leucomalachite green-BSA conjugate on Test line of NC membrane, lead to the color change of Test line. When the sample has no Leucomalachite green residue or concentration lower than detection limit, T line is darker than C line or T line has same color with C line; when the concentration is equal to or higher than detection limit, T line is obviously lighter than C line or T line has no color. No matter whether there is Leucomalachite green residue in sample, C line will appear, it means the test is valid.
6. Materials required but not provided
1) Equipment: Homogenizer, vortex(optional), centrifuge, Micropipette, balance (a reciprocal sensibility of 0.01 g);
2) Consumables: 50ml/10ml/2ml centrifuge tube, disposable tips.
7. Sample preparation
1) Take 3±0.05g tissue sample after removing fat and homogenized into 50ml centrifuge tube, add 1 bottle of Reagent A, then add 7ml reagent B, shake by vortex for 3min, then centrifuge at 4000r/min for 5min;
2) Transfer 3ml up-layer clear liquid into another 10ml centrifuge tube (Note: If the up-layer liquid is less than 3ml, remove all the up-layer liquid and do not take the down-layer liquid), add 100µl reagent C, shake by hand up-and-down for 10S, then add 150μl reagent D, shake by hand up-and-down for 10S, then centrifuge at 4000r/min for 1min (or be static for 5min);
3) Add 170μl reagent E into 2ml centrifuge tube, add 50μl down-layer liquid from step 2, use Micropipette to blow and beat mix it evenly, ready to test.
8. Operation procedures
8.1 Read the instruction carefully before use. Return test cards and sample into room temperature.
8.2 Take out test bag, open and take out required microwells, making proper marks. Please use them within 1h. Seal the test bag, avoid moisture.
8.3 Take 100µl of the test samples into the microwells, then repeatedly pump and suck for many times, mix the sample with the reagent in the microwells completely until no solid judged by eyes (this is a very important step). Incubate for 3min at room temperature (20-25℃).
8.4 Put the test card flatly, absorb all liquid from the micro-well, add into the “S” hole on test card.
8.5 Be static for 5~8min, read the result in 1min according to diagram, it is invalid in other time.
9. Test Result Interpretation
9.1 Negative: T line is darker than C line, or T line has same color with C line. It means there is no Leucomalachite green residue in sample or the residue is lower than detection limit.
9.2 Positive: T line is obviously lighter than C line or T line is invisible or T line become green or blue color. It means the Leucomalachite green residue is equal to or higher than detection limit.
9.3 Invalidation: C line isn’t seen wine red. It means the test card is out of efficacy, out of date or improper operation. Please run the test again using another package. If the invalid tests keep happening, please contact the supplier.
10. Precautions
1) The test card can be used only once at room temperature, do not use test card out of expiry date.
2) Do not touch the white membrane surface in the middle of test card, avoid sunlight and fan blowing directly.
3) Excessive fat content in the sample can easily lead to emulsification, making it impossible to remove the supernatant from step 2. Therefore, it is necessary to select a sample that has been homogenized after removing fat.
4) Due to production process, the T-line may become blue, which is a normal phenomenon and does not affect the result judgment
5) Test again for positive result;

11.Specificity
Test 500ppb Tetracyclines, Quinolones and Sulfonamides etc. The result is Negative.
