2. Brief

This product is used for testing Malachite green or Leucomalachite Green in Tissue (include poultry and livestock muscles, fish, shrimp, crab and other aquatic product muscles) sample qualitatively. Easy to operate, high sensitivity.

5. Principle

The Malachite green rapid test card is based on competitive inhibition immuno-chromatographic principle. In the flow process, Malachite green in the sample combined with Malachite green specific colloidal gold-labeled monoclonal antibody, Inhibit the combination between antibody and Malachite green-BSA conjugate on Test line of NC membrane, lead to the color change of Test line. When the sample has no Malachite green residue or concentration lower than detection limit, T line is darker than C line or T line has same color with C line; when the concentration is equal to or higher than detection limit, T line is obviously lighter than C line or T line has no color. No matter whether there is Malachite green residue in sample, C line will appear, it means the test is valid.

6. Materials required but not provided

1) Equipment: Homogenizer, nitrogen-drying device, vortex(optional), centrifuge, Micropipette, balance (a reciprocal sensibility of 0.01 g);

2) Consumables: 50ml centrifuge tube, disposable tips.

7. Sample preparation

1) Take 3±0.05g homogenized tissue sample into 50ml centrifuge tube, add 2ml Reagent 1, then add 6ml reagent 2, shake by hand or by vortex strongly for 2min (shake by hand up-and-down strongly), add 1 bottle of reagent 3, shake strongly for 2min (shake by hand up-and-down strongly), then centrifuge at 4000r/min for 5min;

2) Transfer 3ml up-layer clear liquid into another centrifuge tube (Note: please do not take down-layer liquid, otherwise the test card may become invalid), add 100µl reagent 4, shake for 30S. Blow to dry at 56℃ by Nitrogen or air(or use hair-dryer);

3) Add 0.5ml reagent 5, shake for 2min (add a 200ul tips into the centrifuge tube) to fully dissolve the residue on wall of centrifuge tube, absorb 120ul down-layer liquid as sample, ready to test.

8. Operation procedures

8.1 Read the instruction carefully before use. Return test cards and sample into room temperature.

8.2 Take out test bag, open and take out required microwells, making proper marks. Please use them within 1h. Seal the test bag, avoid moisture.

8.3 Take 120µl of the test samples into the microwells, then repeatedly pump and suck for many times, mix the sample with the reagent in the microwells completely until no solid judged by eyes (this is a very important step). Incubate for 1-2min at room temperature (20-25℃) (If an invalid test card appears, incubation treatment may not be required.).

8.4 Put the test card flatly, absorb the 80μl liquid from the micro-well, add into the “S” hole on test card.

8.5 Be static for 5~8min, read the result in 1min according to diagram, it is invalid in other time.

9. Test Result Interpretation

9.1 Negative: T line is darker than C line, or T line has same color with C line. It means there is no Malachite green residue in sample or the residue is lower than detection limit.

9.2 Positive: T line is obviously lighter than C line or T line is invisible. It means the Malachite green residue is equal to or higher than detection limit.

9.3 Invalidation: C line isn’t seen wine red. It means the test card is out of efficacy, out of date or improper operation. Please run the test again using another package. If the invalid tests keep happening, please contact the supplier.

10. Precautions

1) The test card can be used only once at room temperature, do not use test card out of expiry date.

2) Disposable tips cannot be reused to avoid cross contamination.

3) Do not touch the white membrane surface in the middle of test card, avoid sunlight and fan blowing directly.

4) Tap water, distilled water, or deionized water cannot be used as negative control.

5) Test again for positive result;

6) Due to the sample differences, some Testing line may appear light or gray in color, but as long as a red band appears, it can be determined.

11. Safety performance and garbage disposal

The test card and its packaging are non-toxic and harmless. After use, dispose of them as dry waste; Tissue sample to be treated as wet waste.

12. Storage and expiry date

Storage: Store at 4-30 ℃ in dark, sealed, dry place, no frozen.

Expiry date: 12 months; date of production is on box.
