[ Components and Contents]

[Function and Purpose]

This kit is used to detect Foot-and-Mouth Disease Virus Serotype SAT-1 antibodies in the serum of infected or immunized animals such as cattle, sheep, and pigs.

[Usage and Determination]

1 Preparation before use

PBST washing solution: dilute the 25x PBST washing solution with deionized water at 25 times(for example: to prepare 500ml PBST solution, need 20ml of 25x PBST washing solution+ 480ml of deionized water.)

2 Operating Steps

1) Take out all components from kit, return them to room temperature(20~25℃) before use.

2) Record the positions of negative and positive control serum, as well as the serum sample, and add the sample to the serum dilution plate. Negative and positive control serum: No dilution is required, add 70 µ l directly to the dilution plate; serum sample: add 63 µ l of sample diluent to each well, then add 7 µ l of serum sample, and gently pipette up and down for 5~8 times to mix it evenly. (Note: each test need to set 2 wells of negative control and 2 wells of positive control serum

• Transfer negative control serum, positive control serum, diluted serum sample into FMD SAT-1 coated plate, 50µl/well, seal the plate with the Adhesive Foil and incubate at 37℃ for 30 minutes.
• Plate washing: remove the liquid from the wells, add 220～300µl of PBST washing solution per well, remove the liquid again, wash 3~5 times, drying the plate with absorbent paper during the final wash.
• Add FMD SAT-1 antibody working solution 50μl /well,seal the plate with the Adhesive Foil and incubate at 37℃ for 30 minutes.
• Washing like step 4).
• Mix Substrate A and Substrate B at 1:1 evenly, prepare the mixed solution for immediately use. Add the mixed substrate solution 50μl /well, and incubate at 37℃ in the dark for 12±1 minutes.
• Add stop solution 50μl /well, then read OD450nm by ELISA reader.
