【Principle and Scope of Application】
Salmonella belongs to the Enterobacteriaceae family, encompassing bacteria that cause food poisoning, gastroenteritis, typhoid, and paratyphoid fever. Foodborne Salmonella infections primarily originate from dairy, poultry, and meat products; chicken and eggs are particularly high-risk foods. The Salmonella test plate contains a selective culture medium, a Salmonella-specific esterase chromogenic indicator, and a high-molecular water-absorbing gel. Utilizing proprietary microbial test plate technology, it functions as a rapid, disposable testing product. The presence of Salmonella can be confirmed with a one-step incubation of 15~24h, making it highly suitable for rapid testing by various inspection departments and food enterprises. This product is applicable for the rapid testing of meat and meat products, eggs and egg products, cold drinks, etc.
【Operating Procedures】
1. Sample Preparation:
Method without enrichment:
① Weigh 25 mL (or 25 g) of the sample and place it into a sterile sampling container or a homogenizer cup containing
225 mL sterile phosphate buffer diluent (or physiological saline). Homogenize to prepare a 1:10 sample suspension.
② Transfer 1 mL of the suspension into 9 mL diluent to prepare a 1:100 dilution.
③ Perform further serial dilutions as required.
Method with enrichment:
① Weigh 25 mL (or 25 g) of the sample and place it into a sterile sampling container or a homogenizer cup containing 225 mL of sterile phosphate buffer diluent (or physiological saline). Homogenize to prepare a 1:10 sample suspension.
② Transfer 1 mL of the suspension into an enrichment broth (prepared separately).
③ Incubate at 36°C ±1°C for 8–18 hours.
④ Use the enriched culture as the first dilution sample.
⑤ Transfer 1 mL of the enriched broth into 10 mL of diluent to prepare the next dilution, and continue serial dilution if necessary.
2. Inoculation:
① Place the Salmonella detection plate on a clean and level laboratory bench.
② Open the lid carefully. Using a sterile pipette, slowly add 1 mL of the sample solution onto the culture medium.
③ While dispensing the sample, gently rotate the plate to ensure the medium is evenly moistened and absorbs the sample completely.
④ For each dilution level, inoculate two plates.
⑤ Allow the plate to stand for 3–5 minutes before incubation. (Note: Rapid dispensing may easily flush awaythe gel powder layer, but subsequent water equilibration will not affect the test results.)
3. Incubation: Place the fully absorbed detection plate upside down in an incubator. Incubation conditions: Temperature: 36°C ±1°C; Time: 15–24 hours.
[Result Interpretation]
Observe the test plates; purple-red colonies indicate Salmonella, while blue colonies indicate other bacterial flora. For samples showing positive colonies, confirmation using more reliable methods is recommended. If confirmation is not available, the test should be repeated with additional samples.
[Reporting of Results]
Report the results as: Salmonella detected / not detected in 25 mL (or 25 g) of the sample.
[Additional Information]
1. Performance Characteristics
Experimental validation shows that pure cultures of Salmonella (including hydrogen sulfide-negative strains) produce typical purple-red colonies on the plate. Other Enterobacteriaceae may produce blue colonies, while Staphylococcus species do not grow on the detection plate.
2. Biosafety Notice
After use, the detection plates may contain viable microorganisms. They must be disposed of promptly according to biosafety waste management regulations.
