1. Brief
This product is used for testing Rhodamine B in Chili products (chili powder, chili oil, chili sauce, etc.) qualitatively. The total test time only needs 30 min. Easy to operate, high sensitivity.

4. Principle
The Rhodamine B rapid test card is based on competitive inhibition immuno-chromatographic principle. In the flow process, Rhodamine B in the sample combined with Rhodamine B specific colloidal gold-labeled monoclonal antibody, Inhibit the combination between antibody and Rhodamine B-BSA conjugate on Test line of NC membrane, lead to the color change of Test line.

6. Materials needed but not provided.
Instruments: electronic balance, centrifuge, micropipette, vortex oscillator, homogenizer.
Consumables: 15/10ml centrifuge tube, disposable tips.
7. Sample preparation
7.1 Chili sauce
1). Accurately weigh 0.5g of sample and add it to a 15ml centrifuge tube.
2). Accurately add 5ml of ethanol to the centrifuge tube, tightly seal the stopper, shake vigorously for 3 minutes, and centrifuge at 4000rpm for 5 minutes.
3). Transfer 4mL of the supernatant after stratification to a 10ml centrifuge tube, and dry it using a nitrogen-blowing instrument at 60℃ (it is normal for some granular orange-yellow chili oil residue to remain undried at the bottom of the centrifuge tube). Add 400μl of the Rhodamine B redissolving solution to the dried 10ml centrifuge tube, shake and mix thoroughly for 30 seconds until it is fully dissolved, and then centrifuge at 4000rpm for 3 minutes. The lower layer is the sample solution for testing (after centrifugation, the upper layer is the oil layer, and the lower layer is the sample solution).
7.2 Chili powder:
1). Accurately weigh 2g of sample and add it to a 15ml centrifuge tube.
2). Accurately add 5ml of ethanol to the centrifuge tube, then add 1.5ml of Rhodamine B extracting solution. Secure the stopper tightly and shake vigorously for 3 minutes. Centrifuge at 4000rpm for 5 minutes.
3). Transfer 3mL of the supernatant (if less than 3mL, transfer the entire amount) from the stratified solution to a 10mL centrifuge tube, add 1 tube of Rhodamine B extractant, shake and mix thoroughly for 1 minute, and centrifuge at 4000rpm for 3 minutes.
4). Transfer 2mL of the supernatant after stratification to a 10ml centrifuge tube, and dry it with a nitrogen blower at 60℃ (it is normal for some granular orange-yellow chili oil residue to remain undried at the bottom of the centrifuge tube). Add 400μl of the Rhodamine B redissolving solution to the dried 10ml centrifuge tube, shake and mix thoroughly for 30 seconds until fully dissolved, and centrifuge at 4000rpm for 3 minutes. 8. Operation procedures
8.1 Read the instruction carefully before use. Return test card and sample into room temperature.
8.2 Take out required micro-wells and test card, making proper marks. Please use it within 1h. Please immediately cover the remaining micro-wells to prevent the remaining reagent from getting wet.
8.3 Take 100µl of the sample solution into the micro-wells, then repeatedly pipette for many times, mix the sample with the reagent in the micro-wells completely until no solid judge by eyes (this is a very important step). Incubate for 5min at room temperature (20-25℃).
8.4 Put test card flatly, absorb all liquid from micro-well, drop it into “S” well on test card.
8.5 React then wait for 5~8min and read the result, it is invalid in other time.
9. Test Result Interpretation
9.1 Negative: C line appears, T line is darker than C line, or T line has same color with C line. It means there is no Rhodamine B residue in sample or the residue is lower than detection limit.
9.2 Positive: C line appears, T line is obviously lighter than C line or T line is invisible. It means the Rhodamine B residue is equal to or higher than detection limit.
9.3 Invalidation: C line isn’t seen wine red. It means the test card is out of efficacy, out of date or improper operation. Please run the test again using another package. If the invalid tests keep happening, please contact the supplier.

10. Specificity
Use this test card to test 500ppb Sudan Red, the result is negative.
11. Precaution
1). Rhodamine B redissolving solution, Rhodamine B extractant and Rhodamine B extracting solution are non-toxic and harmless.
2). The test card is disposable at room temperature; Do not use expired test card.
3). Disposable tips cannot be reused to avoid cross contamination.
4). Try not to touch the white film surface in the center of the test card during use; Avoid direct sunlight and direct fan blowing.
5). Tap water, distilled water, deionized water, or sample extracts cannot be used as negative controls.
6). If a positive result appears, it is recommended to retest with new test card.
7). Due to differences in samples, some testing lines may appear lighter or grayish in color, but as long as a red band appears, it can be determined.
8). Keep the Rhodamine B redissolving solution sealed when not in use.
12. Safety performance and garbage disposal
This product's test card and its packaging are non-toxic and harmless, and should be disposed of as dry waste after use; tissue samples should be disposed of as wet waste; the organic reagents used (some of which are toxic and harmful) should be collected in waste liquid bottles for centralized disposal.
13. Storage and expiry date
Storage: Store at 4-30 ℃ in dark, sealed, dry place, no frozen.
Expiry date: 12 months; date of production is on box.
