Rabies virus (RBV) antibody rapid test kit

The Rabies Virus (RBV) antibody rapid test kit (colloidal gold method) uses blocking immunochromatography technology to detect rabies virus antibodies in the serum of animals such as cats and dogs. The neutralization reaction tube contains rabies virus antigen. After the serum to be tested is added to the neutralization reaction tube and left to stand, if the rabies virus antibody titer in the sample is less than 1 IU/mL, the rabies virus antigen in the reaction tube cannot be completely neutralized. Dilute this mixture and add it to the sampling well. Combine the rabies virus antibody labeled with colloidal gold on the pad with the rabies virus to form an antigen-antibody complex, which moves along the chromatographic membrane together, It binds to the detected rabies virus antibody and displays a wine red color. If the titer of rabies virus antibody reaches or exceeds 1 IU/mL, it can completely neutralize the rabies virus antigen in the reaction tube, and no color reaction occurs.

3. Test Procedure
1). Collect 0.5-1mL of blood and centrifuge at 3000 rpm for 3-5 minutes to separate the serum. It can also be left at 4 ℃ overnight to allow the serum to naturally precipitate.
2). Open the aluminum foil bag, take out the neutralization reaction tube (EP tube), and use a micropipette to take 50 μL serum, add to the bottom of the neutralization reaction tube and gently blow repeatedly for at least 5 times to completely mix the serum with the trace reactants at the bottom of the tube.
3). Leave the neutralization reaction tube containing serum at room temperature for 10 minutes.
4). Use a dropper bottle containing sample buffer to add two drops of sample buffer to the neutralization reaction tube (avoid bubbles, do not add too much buffer), and then use a pipette to blow and mix. Place the test card flat and add all the liquid in the neutralization reaction tube to the sampling hole of the test card.
5) At this point, you will see a wine red liquid flowing through the observation window. After adding the sample, time it for 10-15 minutes to interpret the results. The results after 15 minutes are invalid.
4. Result judgment
1) Negative: Both on T Line and C line being seen wine-red color reaction, it means the antibody titer in the serum to be tested is lower than 1 IU/mL.
2) Positive: no color reaction on Test Line (T Line),only on Control Line(C Line) being seen wine-red color reaction, it means the antibody titer in the serum to be tested is higher than 1 IU/mL.
3) Invalid: In the observation window, there are no color lines in the control line area (C) and the testing line area (T); Or only color lines appear in the testing line area (T)., it means the operation is wrong or the test card is invalid, test again.
5. Precautions
1). Products that have expired or have damaged aluminum foil packaging bags cannot be used.
2). Please use the accompanying neutralization reaction tube and sample buffer. Do not mix buffer solutions between different products or buffer solutions or neutralization reaction tubes from different batches of the same product.
3). The test card must be used within 1 hour after removal.
4). Testing each sample requires replacing the pipette tips. Please ensure that there is no cross contamination between samples during the operation.
5). If the collected serum samples cannot be immediately tested, they should be stored at 4 ℃ for a short period of time (48 hours), and below -20 ℃ for long-term storage.
6). The serum sample must be clarified. If there are any visible particles, centrifuge at 3000 rpm for 10 minutes, transfer the serum supernatant, and discard the precipitate.
7). Excessive addition of samples or excessive dripping of sample buffer can lead to incorrect results.
8). There is no correlation between the effectiveness of the experiment and the depth of the control line (C-line). As long as the control line is clearly visible, it indicates that the experiment is effective.
9). The antigen used in the test card has no potential biological hazards, but all serum samples should be properly disposed of according to local pollutant treatment regulations.
10). This product is only for veterinary diagnostic reference and is not intended for clinical diagnosis in humans.
6. Frequently Asked Questions
1) What are the limitations of this product?
The detection threshold of this test card is 1 IU/ml, but there is no strict linear relationship between the results of the colloidal gold chromatography rapid test method and the results of the virus neutralization experiment. This test is only a qualitative test. If accurate quantification is required, please refer to the national or OIE standard methods.
