Brucella antigen rapid test card

1. Principle
The Brucella antigen rapid test card, using colloidal gold immuno-chromatographic method, to detect the Brucella antigen. After the sample is added to the “S” well, it is moved along the chromatographic membrane together with the colloidal gold-labeled anti-BSP monoclonal antibody. If a BSP antigen is present in the sample, it binds to the antibody on the test line to display wine red. If the BSP antigen is not present in the sample, no color reaction occurs.

3. Test procedure
3.1 Sample preparation
3.1.1 Pure cultured bacteria: Pick a single colony of suspected Brucella, and place it in the sample buffer, cover the tube and shake vigorously for more than 30 seconds, then let it stand for 5 minutes. After the large particles are precipitated, the supernatant is taken as the sample solution.
3.1.2 Clinical specimens: Use cotton swab to obtain Inguinal lymph node acupuncture aspirate, vaginal discharge, and diseased tissue homogenate from sick animals with suspected brucellosis. Immediately insert the cotton swab into the tube containing the sample buffer, rotate the cotton swab repeatedly on the tube wall at least 10 times, and mix the solution to dissolve the sample as much as possible in the sample buffer. Squeeze the cotton swab on the wall of the tube above the liquid level to allow the liquid to be squeezed out as much as possible and discard the cotton swab. Cover the tube and shake it vigorously for more than 30 seconds, then let it stand for 5 minutes. After the large particles are precipitated, the supernatant is taken as the sample solution.
3.1.3 Milk: collect 20ml raw milk, centrifuge at 2000g for 15 minutes, discard the supernatant, use a cotton swab to pick up the Precipitate at bottom of centrifuge tube, then immediately insert the cotton swab into the tube containing the sample buffer. Rotate the cotton swab repeatedly on the test tube wall at least 10 times, and mix the solution to dissolve the sample as much as possible in the sample buffer. Squeeze the cotton swab on the wall of the tube above the liquid level to allow the liquid to be squeezed out as much as possible and discard the cotton swab. Cover the tube and shake it vigorously for more than 30 seconds, then let it stand for 5 minutes. After the large particles are precipitated, the supernatant is taken as the sample solution.
3.2 Take out test card, put it on flat desk. Use dropper to take sample solution, add 5 drops into “S” mark well accurately, slowly drop by drop.
3.3 Put it at room temperature for 15~20mins and read the result. It is invalid after 20mins.
4. Results judgment
1) Positive: control line and test line are both seen wine red;
2) Negative: only control line is seen wine red;
3) Invalidation: control line isn’t seen wine red.
5. Storage and expiry date
Store at 2 - 30ºC and the shelf life is 24 months.
6.Notice
1) Do not use product with broken Aluminum foil bag package. Use test card in 1 hour after taking out.
2) The collected sample should be tested immediately, avoid long-tern storage.
3) The sample collection directly affects the accuracy of the test, so ensure that the sample is collected correctly and adequately.
4) Avoid bubble when adding sample.
5) Treat the test card and sample as pollutant according to local regulations.
6) Do not use test card out of date.
