5. Principle
TheNifursol metabolite rapid test kit applies the principle of competitive inhibition immunochromatography. When the sample solution is dropped into the sample hole, theNifursol metabolite in the sample solution combines with the colloidal gold labeled antibody, which then blocks the antigen-binding site of thenifursol on the colloidal gold labeled antibody and prevents the colloidal gold labeled antibody from binding with theNifursol metabolite protein conjugate on the cellulose membrane. When the content of thenifursol metabolites in the sample was higher than or equal to the detection limit, no red line appeared in the detection line (T-line) or the color was weaker than that in line C, and the result was positive. On the contrary, when there is no thenifursol metabolite in the sample or the content is lower than the detection limit, the color rendering of the detection line (T-line) is deeper than line C or consistent with line C, and the result is negative.
6. Sample preparation
Tissue:
1) Take 5±0.05g homogenized tissue sample into 50ml centrifuge tube, add 4.5ml deionized water,1.5ml 1M HCl and 300ul Derivatization reagent, shake and mix for 2min, put at 75℃ water bath for 5min;
2) Add 7ml 1M K2HPO4 solution, shake and mix for 1min, add 6ml ethyl acetate, shake and mix for 1min, centrifuge at 4000r/min for 5min;
3) Take 3ml up-layer liquid into 10ml centrifuge tube, blow to dry at 75℃ by Nitrogen or air.
4) Add 500μl sample dilution, shake to even for 30s, then add 1ml n-hexane, shake up-and-down evenly for 10 times. Centrifuge at 4000r/min for 5min;
5) Absorb 100ul down-layer liquid, ready to test
7. Operation procedures
7.1 Read the instruction carefully before use. Return test strips and sample into room temperature.
7.2 Take out required microwells and strips, making proper marks. Please use these test strips within 1h. Seal the cap of the tubes, avoid moisture.
7.3 Take 200ul of the test samples into the microwells, then slow suction for 5 times, mix the sample with the reagent in the microwells completely until no solid judged by eyes (this is a very important step).
7.4 Incubate for 3min at room temperature (20-25℃), insert the test strips into the microwells with the "MAX" end fully dipped in to the mixture solution.
7.5 Incubate for 3-5min at room temperature again, read the result within 5 minutes, it is invalid in other time.
8. Test Result Interpretation
8.1 Negative: T line is darker than C line, or T line has same color with C line. It means there is no theNifursol metabolite residue in sample or the residue is lower than detection limit.
8.2 Positive: T line is obviously lighter than C line or T line is invisible. It means theNifursol metabolite residue is equal to or higher than detection limit.
8.3 Invalidation: C line isn’t seen wine red. It means the test card is out of efficacy, out of date or improper operation. Please run the test again using another package. If the invalid tests keep happening, please contact the supplier.
9. Precautions
1) The test strip can be used only once at room temperature, do not use test strip out of expiry date.
2) Do not repeat use the tips to avoid cross pollution.
3) Do not touch the white membrane surface in the middle of test strip, avoid sunlight and fan blowing directly.
4) Wear gloves when testing.
5) Please contact the supplier for any questions.
10.Specificity
There is no cross reaction with Quinolones, sulfonamides and thechloramphenicol.
11. Storage and expiry date
Storage: Store at 2-8 ℃ in dark, sealed, dry place, no frozen.
Expiry date: 12 months; date of production is on box.
