Swine Mycoplasma Pneumonia antibody ELISA kit

1. Usage and principle
This kit use indirect enzyme-linked immunoassay (ELISA) to detect specific antibody against Mycoplasma in pig serum or plasma qualitatively, for monitoring antibody after pig immune and auxiliary examination of this disease.
If there is Mycoplasma antibody in sample, it will combine with coated Mycoplasma antigen on plate and the added anti-pig enzyme conjugate specific antibody at second step. The indirect combination and substrate react lead to coloration can reflect whether there is protective mycoplasma antibody in sample.

3. Materials required but not provided
ELISA Reader including 450nm and 630nm wavelength, 37℃ Incubator, measuring pipettes.
4. Sample requirement
1) This kit only used to testing pig serum or plasma.
2) To obtain best laboratory result, avoid using sample with NaN3,high cholesterol,bacteria and severe hemolytic.
3) Store serum sample at 2~8℃ for a short term(3 days); store at -20℃ or below -20℃ for a long time, avoid repeated freezing and thawing.
5. Procedure
1). Washing buffer preparation: dilute 10X washing solution with distilled water or deionized water at 1:9 (such as 100ml 10X washing solution + 900ml distilled water or deionized water), mix thoroughly to obtain the washing solution.
2). Sample dilution and adding sample: Dilute serum sample with sample diluent at 1:99 (such as 5ul serum sample + 495ul sample diluent), mix thoroughly, add the diluted serum sample to sample wells, 100ul/well.
If use Serum diluent plate to dilute sample, the method is as following:
Dilute at 1:100: firstly, add sample diluent into sample wells on Mp-Ag Coated plates, 50ul/well. Add 245ul sample diluent into Serum diluent plate, then add 5ul serum, use pipettes blow and beat evenly, then take 50ul into Mp-Ag Coated plates for react.
(Note: Serum diluent plate is disposable, do not repeat use)
3). Adding controls: Negative control and positive control do not need dilute, add directly.
For each test, set 2 wells for negative control, add negative control 100ul /well, 2 wells for positive control, add positive control 100ul /well. Cover plate with adhesive foil, Incubate at 37 ℃ in dark for 30 min.
4). Pour the liquid out of the wells, add diluted washing solution to each well fully. Use the diluted washing solution to wash for 3 times, each time stand for 1 min, at last time pat to dry on absorbent paper.
5). Adding enzyme conjugate: add 50ul enzyme conjugate to each well. Cover plate with adhesive foil, Incubate at 37 ℃ in dark for 30 min, pour the liquid out of the wells. Wash as last step, pat to dry on absorbent paper.
6). Coloration: Measure as required, take equal volume of Substrate A and Substrate B mix evenly, then add the mixture 100ul/well. Cover plate with adhesive foil, Incubate at 37 ℃ in dark for 10 min.
7). Add 50ul Stop solution to stop reaction. Read the OD value with microplate reader at 450nm (Reference-wavelength: 630 nm).
6. ELISA analysis
1) Cut-off value (COV)=Average OD value of Negative control X 2.1 (Negative control OD value< 0.08, calculate as 0.08)
2) Valid determination:
OD Value of Negative control ≤0.20 (the test is invalid when >0.20)
OD Value of Positive control ≥0.50 (the test is invalid when <0.50)
3) Result judgment:
Sample OD value ≥ COV, swine mycoplasma is positive;
Sample OD value < COV, swine mycoplasma is negative.
7. Limitation
This kit is for screening only, not as a basis for diagnosis.
8. Note
1) Return to room temperature before use. Seal the unused micro-well strip and store at 2~8 ℃.
2) Mix reagent evenly before adding sample.
3) Before incubation, seal the plate with adhesive film, do not repeat use adhesive film.
4) Tighten all caps after use, don’t mix caps. Never mix up reagents and manual from test kits in different batch number.
5) The wasted kits and sample should be treated as infection agent. The stop solution is corrosive, be careful to use.

Expiry date: 12 months.
Storage: Store at 2~8℃ in dark, no frozen.
