Instructions for Use
[Product Introduction]
This product is intended for the qualitative detection of aflatoxin B1 in spices. The entire test procedure requires approximately 15–20 minutes. It is easy to operate and offers high sensitivity.

[Test Principle]
The Aflatoxin B1 Rapid Test Card is based on the princi- ple of competitive inhibition immunochromatography. During migration, aflatoxin B1 in the sample binds to colloidal gold-labeled specific anti-aflatoxin B1 mono- clonal antibodies, thereby inhibiting the binding be- tween the antibodies and the aflatoxin B1-BSA conju- gate immobilized on the test line of the NC membrane. This results in a change in the color intensity of the test line.
[Required Equipment and Consumables]
Equipment: electronic balance, centrifuge, micropipette, vorex mixer (optional).
Consumables: 10 mL centrifuge tubes, 2 mL/1.5 mL centrifuge tubes, disposable pipette tips, and purified water.
[Sample Preparation]
Weigh 1 ± 0.05 g of homogenized sample passed through a 20-mesh sieve into a 10 mL centrifuge tube.
Add 4 mL of Sample Extraction Solution to the centri- fuge tube and shake thoroughly for 3 minutes. Centri- fuge at 4000 r/min for 5 minutes to obtain the superna- tant.

Add the supernatant and Sample Diluent according to the following method. Mix well and use for testing.

[Test Procedure]
1. Read the Instructions for Use carefully before use. Bring the test card and the sample to be tested to room temperature before testing.
2. Take out the required number of microwells and test cards, and mark them properly. Use them as soon as possible within 1 hour. Seal the remaining reagents im- mediately to prevent moisture exposure.
3. Using a micropipette, add 200 μL of the prepared sample into the microwell. Slowly aspirate and dispense several times to fully mix the sample with the lyo - philized powder in the microwell. (The Key Step)
4. Incubate at room temperature 20–25°C for 5 minutes. Place the test card on a flat surface, then transfer 100 μL of liquid from the microwell into the sample well.
5. Allow the reaction to proceed for 5 minutes, then ob- serve the result. Interpret the result within 1 minute according to the reference diagram. Results read at any other time are invalid.
[Interpretation of Results]
1. Negative: The color intensity of the T line is stronger than that of the C line, or there is no obvious difference between the T line and the C line. This indicates that the sample does not contain aflatoxin B1, or that its concentration is below the detection limit.
2. Positive: The color intensity of the T line is signifi- cantly weaker than that of the C line, or the T line does not appear. This indicates that the concentration of aflatoxin B1 in the sample is equal to or higher than the detection limit.
3. Invalid: If the C line does not appear, the result is invalid regardless of whether the T line appears. This indicates that the test card may have failed, expired, or that the test was performed improperly. Repeat the test with a new test card. If the issue persists, please contact our company.

[Precautions]
1. The test card is for single use at room temperature. Do not use expired test cards.
2. Disposable pipette tips must not be reused, in order to avoid cross-contamination.
3. Avoid touching the white membrane area in the center of the test card during use. Avoid direct sunlight and direct airflow from fans.
4. Tap water, distilled water, or deionized water must not be used as negative controls.
5. If a positive result is obtained, it is recommended to repeat the test once using this test card.
6. Due to differences between samples, some test lines may appear light or grayish. However, as long as a red band appears, the result may be interpreted accordingly.
7. Due to the complex composition of spice samples, they may contain substances that interfere with the test. Results may differ slightly between negative spiked samples and actual samples due to different tox - in-binding patterns.
8. For highly pigmented or complex spice matrices such as turmeric and other mixed spices, the detection limit may be higher than that of other spice matrices.

[Safety and Waste Disposal]
The test card and its packaging are non-toxic and harmless. Dispose of them as dry waste after use.
[Storage Conditions and Shelf Life]
1. Store sealed at 2–8°C in a dry place protected from light. Do not freeze.
2. Shelf life: 12 months.
