Aflatoxin M1 rapid test strips
(0.025ppb: Sample pre-treatment by Immunomagnetic beads separation techniques)
Catalog No. : GF08Aj5
1. Brief
This product is used for testing Aflatoxin M1 in milk sample qualitatively. The whole test only need 10~20min, easy to operate with high sensitivity.
2. Packing: 96tests/box
3. Components
1) 12 tubes: each tube includes 8 strips and 8 micro-wells red solid reagent;
2) Magnetic bead suspension: 1 bottle (30mL);
3) Eluent: 1 bottle (50mL);
4) Centrifuge tube: 1 pack (192 pieces);
5) Manual: 1 piece.
4. Detection limit
1) Method 1 (direct detection) Detection limit: 0.05 μg/kg
2) Method 2 (immunomagnetic bead method pretreatment) Detection limit: 0.025 μg/kg
5. Applicable sample
Fresh milk, pasteurized milk, pure milk, whole milk powder (dissolve 1g of milk powder in 9ml of pure water and mix it well)
6. Principle
The colloidal gold immunochromatography was used to detect the aflatoxins M1 residue in the samples. The binding of aflatoxins M1 to the specific monoclonal antibody labeled with colloidal gold inhibited the binding of the antibody to the conjugates of aflatoxins M1-BSA on the NC membrane Testing line, which resulted in the color depth variation of the testing line.
7. Testing equipment
1 incubator; 1 Reader (optional); Dry thermostat (100℃,2mL); Magnetic rack (2mL); Vortex meter; Micropipette; timer
8. Operation procedures
8.1 Method 1:
1) Restore the test strip and sample to room temperature before use.
2) Use a micropipette to pipette 200 µ l of the sample into the micro-well, slowly aspirate 5 times until the mixture is uniform, and observe with the naked eye that there is no solid substance.
3) After incubating with a incubator (40±2℃) for 5 minutes, insert the test strip into the micro-well (with the MAX line end facing downwards, allowing it to be fully immersed in the solution).
4) After immersing the test strip in the micro-well for 6 minutes, interpret the results according to the schematic diagram. Please interpret the results within 5 minutes, otherwise interpretation will be invalid.
8.2 Method 2:
1) Restore the sample to room temperature, take out the required quantity of 2mL centrifuge tubes and place them on the centrifuge tube rack. Take 300μl of the magnetic bead suspension which is uniformly shaken in advance to the centrifuge tube;
2) Add 500μL milk sample, thoroughly mix up and down for 3 minutes;
3) Insert the centrifuge tube into the magnetic rack, Magnetic adsorption for 2 minutes. Carefully discard all liquid from the inner cover and tube body to avoid touching the magnetic beads (if there is residual liquid from the magnetic beads on the inner cover, keep the centrifuge tube on the magnetic rack and invert it 2-3 times to fully absorb the magnetic beads);
4) Add 500μL of Eluent immediately, Vortex for 10 seconds or invert it thoroughly until the magnetic beads are completely dispersed in the liquid phase. Transfer all the suspension to a new 2 mL centrifuge tube, and place the centrifuge tube on a dry thermostat at 100℃ for 5 minutes;
5) Insert the centrifuge tube into the magnetic rack, Magnetic adsorption for about 2 minutes. After it recovers to about 30-50℃, take the purified sample solution on the magnetic rack for testing.
6) Use a micropipette to pipette 200 µl of the sample solution into the micro-wells, slowly aspirate 5 times until the mixture is uniform, and observe with the naked eye that there is no solid substance.
7) After incubating with a incubator (40±2℃) for 5 minutes, insert the test strip into the micro-wells(with the MAX line end facing downwards, allowing it to be fully immersed in the solution).
8) After immersing the test strip in the micro-wells for 6 minutes, interpret the results according to the schematic diagram. Please interpret the results within 5 minutes, otherwise interpretation will be invalid
9. Test Result Interpretation
Color depth comparison of T line and C line | Result judgment | Result analysis |
T line > C line | Negative | Aflatoxins M1 residue is lower than detection limit in sample |
T line = C line | Weak positive | Aflatoxins M1 residue is equal to detection limit in sample |
T line < C line or T line is invisible | Positive | Aflatoxins M1 residue is higher than detection limit in sample |
C line is invisible | Invalid | The test dipstick is invalid, test again |
10. Precautions
1) The test strip can be used only once at room temperature, do not use test strips out of expiry date.
2) Please seal the tube after taking out required test strips. If can not use all 8 microwells at once, cup the rest and put it back to tube and sealed.
3) Do not touch the white membrane surface in the middle of test strip, avoid sunlight and fan blowing directly.
4) Milk samples should be fully liquid without any agglomeration and deposition.
5) Use the test strips testing again for positive results.
6) For milk samples with high fat content, the chromatographic speed will be relatively slow, so it is recommended to extend the reaction time of the second step by 1min.
7) The results is for reference only, for confirmation, please refer to the relevant national standard methods.
Test 500μg/kg Sulfonamides, quinolones and tetracyclines and other drugs. All the results are negative.
12. Storage and expiry date
Storage: Store at 2-8 ℃ in dark, sealed, dry place, no frozen.
