5. Principle
The Malachite green rapid test card is based on competitive inhibition immuno-chromatographic principle. In the flow process, Malachite green in the sample combined with Malachite green specific colloidal gold-labeled monoclonal antibody, Inhibit the combination between antibody and Malachite green-BSA conjugate on Test line of NC membrane, lead to the color change of Test line. When the sample has no Malachite green residue or concentration lower than detection limit, T line is darker than C line or T line has same color with C line; when the concentration is equal to or higher than detection limit, T line is obviously lighter than C line or T line has no color. No matter whether there is Malachite green residue in sample, C line will appear, it means the test is valid.
6. Materials required but not provided
1) Equipments: Homogenizer, nitrogen-drying device, vortex(optional), centrifuge, Micropipette, balance (a reciprocal sensibility of 0.01 g);
2) Consumables: 15ml centrifuge tube, disposable tips.
7. Sample preparation
Water sample:
No treatment is required (if there are too many impurities, it needs to centrifuge at 4000 rpm for 5 minutes, or it needs to be left standing). Take 1.8ml water sample, add 0.2ml reagent 1, and mix well for testing
8. Operation procedures
8.1 Read the instruction carefully before use. Return test cards and sample into room temperature.
8.2 Take out test bag, open and take out required micro-wells, making proper marks. Please use them within 1h. Seal the test bag, avoid moisture.
8.3 Take 120ul of the test samples into the micro-wells, then repeatedly pump and suck for many times, mix the sample with the reagent in the micro-wells completely until no solid judged by eyes (this is a very important step). Incubate for 1-2min at room temperature (20-25℃) (if appear invalid test card, then do not need incubation).
8.4 Put the test card flatly, absorb the 80ul liquid from the micro-well, add into the “S” hole on test card.
8.5 Be static, read the result in 5~8min, it is invalid in other time.
9. Test Result Interpretation
9.1 Negative: T line is darker than C line, or T line has same color with C line. It means there is no Malachite green residue in sample or the residue is lower than detection limit.
9.2 Positive: T line is obviously lighter than C line or T line is invisible or T line appear green or blue color. It means the Malachite green residue is equal to or higher than detection limit.
9.3 Invalidation: C line isn’t seen wine red. It means the test card is out of efficacy, out of date or improper operation. Please run the test again using another package. If the invalid tests keep happening, please contact the supplier.
10. Precautions
1) The test card can be used only once at room temperature, do not use test card out of expiry date.
2) Do not touch the white membrane surface in the middle of test card, avoid sunlight and fan blowing directly.
3) Test again for positive result;

11.Specificity
Test 500ppb Tetracyclines, Quinolones and Sulfonamides etc. The result is Negative.
12. Storage and expiry date
Storage: Store at 2-8 ℃ in dark, sealed, dry place, no frozen.
Expiry date: 12 months; date of production is on box.
